Cloning without E. coli — get to your results faster
Get to your results faster and in fewer steps with the Gibson Assembly® RapidAMP™ isothermal amplification. Assemble and amplify transfection-ready DNA in a simple two-day workflow. Achieve up to 10 µg of high-quality, high-fidelity DNA with RapidAMP™ technology, and skip the tedious tasks associated with transformation, cell culture, and E. coli harvest.
A robust amplification workflow
Begin with a linearized vector, plus up to three linear DNA insert fragments — 300 bp to 1,800 bp each — and assemble via Gibson Assembly® technology to generate a circular cloning product.
This representative gel shows clean bands, post-restriction digest and before any DNA purification. After cloning and amplification on the bench, or with the BioXp® system, the final DNA product can be digested to linear form. It is then ready for downstream applications, negating the need for traditional E. coli transformation and plasmid purification. RapidAMP™ technology has been used successfully for transfection — lipofection and electroporation — and in vitro transcription. The Gibson Assembly® RapidAMP™ workflow produces clean, high-yield DNA material in only a few hours.

Scale up with the BioXp™ system
Revolutionizing synthetic biology
Gibson Assembly® RapidAMP™ reagents are available as a benchtop reagent kit or an automated solution for the BioXp™ system. The BioXp™ system enables up to 24 constructs to be built, cloned, and amplified in a single overnight run, eliminating the need to transform into E. coli.
The BioXp™ system starts by synthesizing the required fragments — up to 24 at a time — and then cloning them into up to 4 vectors of interest. By the following afternoon, these clones are amplified up to 10 µg DNA using Gibson Assembly® RapidAMP™ technology.
